地黄叶多糖的超高压提取工艺优化及抗氧化活性研究

摘要:为充分利用地黄叶资源,采用超高压提取方法提取地黄叶中多糖,并且将其与热水提取、微波提取和超声波提取的地黄叶多糖进行抗氧化活性的比较。结果表明:超高压压力400 MPa、加压时间8.5 min、液固比20∶1 mL/g为最佳提取条件,在该条件下,地黄叶中多糖得率达到4.52%,其对DPPH自由基和ABTS+自由基清除能力的IC50分别为1.26 mg/mL1.78 mg/mL,清除能力强于热水提取、微波提取和超声波提取的地黄叶多糖。超高压提取方法提取地黄叶多糖得率高、时间短、活性高。在提取地黄叶多糖方面具有较好的应用前景。

Optimization of Ultra-high Pressure Extraction and Antioxidant Activity of Polysaccharides from Rehmannia glutinosa Libosch Leaves

AbstractIn order to make full use of leaves of Rehmannia glutinosa Libosch, ultra-high pressure extraction was applied to extract of polysaccharide from leaves. The extracted polysaccharides was compared with those from hot water extraction,microwave and ultrasonic extraction. The optimal extraction parameters to obtain the highest polysaccharide yield were as follows: pressure 400 MPa, extraction time 8.5 min and liquid-solid ratio 20∶1 mL/g. The yield of polysaccharide under the optimum conditions was found to be 4.52%. The IC50 of DPPH radical scavenging activity and ABTS radical scavenging activity are 1.26 mg/mL and 1.78 mg/mL, respectively. It was better than that of hot water extraction, microwave extraction and ultrasonic extraction. Compared with hot water extraction, microwave extraction and ultrasonic extraction, ultra- high pressure extraction has higher extraction rate, shorter extraction time and higher activity. It has a good application prospect in extracting polysaccharides from Rehmannia glutinosa leaves.
